Ipamorelin is a selective growth-hormone secretagogue that binds to the growth-hormone secretagogue receptor type 1a (GHS-R1a) without significant activity at ghrelin O-acyltransferase or other pituitary hormone receptors. In in vitro and preclinical rodent models, it’s employed to dissect growth-hormone axis dynamics and downstream anabolic, lipolytic, and osteoanabolic pathways.

          Mechanism of action

          GHS-R1a Agonism & GH Secretion

          Ipamorelin selectively activates GHS-R1a on somatotrophs, coupling via Gαq/11 to PLCβ → IP₃/DAG signaling, triggering Ca²⁺-dependent exocytosis of growth hormone—yielding pulsatile release without concomitant ACTH or cortisol secretion.

          Anabolic & Lipolytic Signaling

          Elevated GH induces hepatic IGF-1 production, which engages the IGF-1R/PI3K/Akt pathway in muscle and adipose tissue, promoting protein synthesis and adipocyte lipolysis.

          Osteoanabolic Effects

          GH/IGF-1 axis activation upregulates Runx2 and osterix in osteoblasts via JAK2/STAT5 signaling, enhancing bone matrix deposition and mineralization.
