AOD-9604 is a synthetic hexadecapeptide fragment corresponding to residues 176–191 of human growth hormone. It selectively stimulates adipocyte lipolysis by upregulating β₃-adrenergic receptors and inhibiting lipoprotein lipase, while sparing IGF-1 receptor activation. The peptide demonstrates oral bioavailability and reduces weight gain in rodent models without altering blood glucose levels.

          Mechanism of action

          β₃-Adrenergic Receptor–Mediated Lipolysis

          AOD-9604 upregulates β₃-adrenergic receptor expression in adipocytes, increasing intracellular cAMP and activating hormone-sensitive lipase for fatty-acid mobilization

          Lipoprotein Lipase Inhibition

          The peptide directly inhibits adipose tissue lipoprotein lipase activity, shifting the balance toward enhanced fat breakdown over storage.

          Oral Bioavailability

          In obese Zucker rats, once-daily oral dosing (500 µg/kg) achieved systemic exposure and a >50% reduction in weight gain, demonstrating effective gastrointestinal absorption
